Drug Resistance and R-plasmid of Gram Negative Bacterial to Perfilae Herba Extracts and Antimicrobial Agents

Abstract
Forty-eight strains of @ Salmonella and forty-two strains of Serratia species isolated from patients and twenty-one strains of Vibrio species isolated from coastal areas in Mokpo were tested for their susceptibility to Perillae herba extracts and fourteen antimicrobial agents. The transfer of drug resistance was tested by conjugation. The following results were obtained. 1. Aqueous extract and ethanol extract of Perillae herbs was the most active against Vibrio. Aqueous extract was more effective than ethanol extract. 2. The frequency of resistant straifn of Salmonella was the highest to streptornycin(Sm) with 62.5%, Serratia to cephradine(Cr) with 100%, and Vibrio to cephradine(Cr) with 71.43%. And 31 strains (64.58%) of Salmonella species, 42 strains (100%) of Serratia species and 16 strains (76.19) of Vibrio species were resistant to one or more drugs tested. The frequency of multiple drug-resistant strains was much higher in the Serratia species than in the Salmonella or Vibrio species. 3. Minimum inhibitory concentration (MIC) of third-generation cephalosporins, aztreonam and ciprofloxacin was very low compared with those of other antimicrobial agents tested, and Salmonella showed a tendency to be more sensitive than Serratia or Vibrio. 4. In the transfer test of drug resistance by conjugation, multiple drug-resistant strains transferred their partial.pattems of resistance to recipient and the transfer was considered to be mediated by R-plasmid. 5. The plasn-iid profiles were studied by the methods of SDS lysis and agarose gel electrophoresis. Multiple drug- resistant strains of Salmenella showed one or two plas@ds of molecular size ranging 3.50 to 26.55 Mdal. The conjugative R-plasmids of 11.63 Mdal and 26.55 Mdal were noted in transconjugants. In the multiple drug-resistant strains of Seffatia, one to three plasrnids of 5.09-35.8 Mdal were transferred by conjugation.
